    NMR and Electron Microscopy Studies of Soybean Seeds, Soybean Proteins, and Oil for Improved Utilization in Foods and Feeds

    NMR, NIR and EM techniques are suitable methods of measuring protein and oil content in soybeans. After completion of a measurement with NMR or NIR, a soybean sample could be either replanted or consummated. NMR techniques involved in the present work are based on the response of nuclei, as nuclear spin flips, when set in a 7.05T magnetic field and excited with rf pulses of 300 MHz. NIR spectroscopy is based on outer electron molecular vibrations, when excited with radiation of frequency 1012 -4×1014Hz, and =2.5Om-750 nm. 
TEM and ESEM techniques are bulk and surface analysis techniques, respectively, working with electron beams of 15KV, and image resolution in the range of nm’s. ESEM is based on the possibility of imaging with secondary emitted electrons from the samples, and TEM on the possibility to have image contrast based on the difference in electron transmission through elements of different atomic weight. Novel results are presented by NMR, FT-NIR spectroscopy and Electron Microscopy for intact, selected soybean seeds accessions, soybean oil and soybean proteins

    NIR Calibrations for Soybean Seeds and Soy Food Composition Analysis: Total Carbohydrates, Oil, Proteins and Water Contents [v.2]

    Conventional chemical analysis techniques are expensive, time consuming, and often destructive. The non-invasive Near Infrared (NIR) technology was introduced over the last decades for wide-scale, inexpensive chemical analysis of food and crop seed composition (see Williams and Norris, 1987; Wilcox and Cavins, 1995; Buning and Diller, 2000 for reviews of the NIR technique development stage prior to 1998, when Diode Arrays were introduced to NIR). NIR spectroscopic measurements obey Lambert and Beer’s law, and quantitative measurements can be successfully made with high speed and ease of operation. NIR has been used in a great variety of food applications. General applications of products analyzed come from all sectors of the food industry including meats, grains, and dairy products (Shadow, 1998).
Novel NIR calibrations for rapid, reliable and accurate composition analysis of a variety of several soy based foods and bulk soybean seeds were developed and validated in a six-year collaborative project with a large number of different samples (N >~12, 000). The availability of such calibrations is important for establishing NIR as a secondary method for composition analysis of foods and soybeans both in applications and fundamental research

    Determination of Soybean Oil, Protein and Amino Acid Residues in Soybean Seeds by High Resolution Nuclear Magnetic Resonance (NMRS) and Near Infrared (NIRS)

    A detailed account is presented of our high resolution nuclear magnetic resonance (HR-NMR) and near infrared (NIR) calibration models, methodologies and validation procedures, together with a large number of composition analyses for soybean seeds. NIR calibrations were developed based on both HR-NMR and analytical chemistry reference data for oil and twelve amino acid residues in mature soybeans and soybean embryos. This is our first report of HR-NMR determinations of amino acid profiles of proteins from whole soybean seeds, without protein extraction from the seed. It was found that the best results for both oil and protein calibrations were obtained with a Partial Least Squares Regression (PLS-1) analysis of our extensive NIR spectral data, acquired with either a DA7000 Dual Diode Array (Si and InGaAs detectors) instrument or with several Fourier Transform NIR (FT-NIR) spectrometers equipped with an integrating sphere/InGaAs detector accessory. In order to extend the bulk soybean samples calibration models to the analysis of single soybean seeds, we have analized in detail the component NIR spectra of all major soybean constituents through spectral deconvolutions for bulk, single and powdered soybean seeds. Baseline variations and light scattering effects in the NIR spectra were corrected, respectively, by calculating the first-order derivatives of the spectra and the Multiplicative Scattering Correction (MSC). The single soybean seed NIR spectra are broadly similar to those of bulk whole soybeans, with the exception of minor peaks in single soybean NIR spectra in the region from 950 to 1,000 nm. Based on previous experience with bulk soybean NIR calibrations, the PLS-1 calibration model was selected for protein, oil and moisture calibrations that we developed for single soybean seed analysis. In order to improve the reliability and robustness of our calibrations with the PLS-1 model we employed standard samples with a wide range of soybean constituent compositions: from 34% to 55% for protein, from 11% to 22% for oil and from 2% to 16% for moisture. Such calibrations are characterized by low standard errors and high degrees of correlation for all major soybean constituents. Morever, we obtained highly resolved NIR chemical images for selected regions of mature soybean embryos that allow for the quantitation of oil and protein components. Recent developments in high-resolution FT-NIR microspectroscopy extend the NIR sensitivity range to the picogram level, with submicron spatial resolution in the component distribution throughout intact soybean seeds and embryos. Such developments are potentially important for biotechnology applications that require rapid and ultra- sensitive analyses, such as those concerned with high-content microarrays in Genomics and Proteomics research. Other important applications of FT-NIR microspectroscopy are envisaged in biomedical research aimed at cancer prevention, the early detection of tumors by NIR-fluorescence, and identification of single cancer cells, or single virus particles in vivo by super-resolution microscopy/ microspectroscopy

mature soybean embryos by FT-IR/NIR Microspectroscopy. This is the first report of FT-NIR Chemical Images obtained with 1 micron resolution. NIR spectra of Proteins, Oil and DNA fibers are obtained for regions as small as 1μ^2^

    Requirements for Robotic Interpretation of Social Signals “in the Wild”: Insights from Diagnostic Criteria of Autism Spectrum Disorder

    The last few decades have seen widespread advances in technological means to characterise observable aspects of human behaviour such as gaze or posture. Among others, these developments have also led to significant advances in social robotics. At the same time, however, social robots are still largely evaluated in idealised or laboratory conditions, and it remains unclear whether the technological progress is sufficient to let such robots move “into the wild”. In this paper, we characterise the problems that a social robot in the real world may face, and review the technological state of the art in terms of addressing these. We do this by considering what it would entail to automate the diagnosis of Autism Spectrum Disorder (ASD). Just as for social robotics, ASD diagnosis fundamentally requires the ability to characterise human behaviour from observable aspects. However, therapists provide clear criteria regarding what to look for. As such, ASD diagnosis is a situation that is both relevant to real-world social robotics and comes with clear metrics. Overall, we demonstrate that even with relatively clear therapist-provided criteria and current technological progress, the need to interpret covert behaviour cannot yet be fully addressed. Our discussions have clear implications for ASD diagnosis, but also for social robotics more generally. For ASD diagnosis, we provide a classification of criteria based on whether or not they depend on covert information and highlight present-day possibilities for supporting therapists in diagnosis through technological means. For social robotics, we highlight the fundamental role of covert behaviour, show that the current state-of-the-art is unable to characterise this, and emphasise that future research should tackle this explicitly in realistic settings

    Ficaria verna Huds. extracts and their β-cyclodextrin supramolecular systems

    <p>Abstract</p> <p>Background</p> <p>Obtaining new pharmaceutical materials with enhanced properties by using natural compounds and environment-friendly methods is a continuous goal for scientists. <it>Ficaria verna </it>Huds. is a widespread perennial plant with applications in the treat of haemorrhoids and to cure piles; it has also anti-inflammatory, astringent, and antibiotic properties. The goal of the present study is the obtaining and characterization of new <it>F. verna </it>extract/β-cyclodextrin complexes by using only natural compounds, solvents, and environment-friendly methods in order to increase the quality and acceptability versus toxicity indicator. Thus, the flavonoid content (as quercetin) of <it>Ficaria verna </it>Huds. flowers and leaves from the West side of Romania was determined and correlated with their antioxidant activity. Further, the possibility of obtaining β-cyclodextrin supramolecular systems was studied.</p> <p>Results</p> <p><it>F. verna </it>flowers and leaves extracts were obtained by semi-continuous solid-liquid extraction. The raw concentrated extract was spectrophotometrically analyzed in order to quantify the flavonoids from plant parts and to evaluate the antioxidant activity of these extracts. The <it>F. verna </it>extracts were used for obtaining β-cyclodextrin complexes; these were analyzed by scanning electron microscopy and Karl Fischer water titration; spectrophotometry was used in order to quantifying the flavonoids and evaluates the antioxidant activity. A higher concentration of flavonoids of 0.5% was determined in complexes obtained by crystallisation method, while only a half of this value was calculated for kneading method. The antioxidant activity of these complexes was correlated with the flavonoid content and this parameter reveals possible controlled release properties.</p> <p>Conclusions</p> <p>The flavonoid content of <it>F. verna </it>Huds. from the West side of Romania (Banat county) is approximately the same in flowers and leaves, being situated at a medium value among other studies. β-Cyclodextrin complexes of <it>F. verna </it>extracts are obtained with lower yields by crystallisation than kneading methods, but the flavonoids (as quercetin) are better encapsulated in the first case most probably due to the possibility to attain the <it>host</it>-<it>guest </it>equilibrium in the slower crystallisation process. <it>F. verna </it>extracts and their β-cyclodextrin complexes have antioxidant activity even at very low concentrations and could be used in proper and valuable pharmaceutical formulations with enhanced bioactivity.</p
